Abstract:

Molecular studies investigating phylogeographical issues are tending to focus on the control region of the mitochondrial genome (also named D-loop). Several factors, including the mitochondria’s maternal inheritance pattern, its lack of recombination and its relatively rapid evolution (as compared to the nuclear genome) have made the control region the ideal locus for examining complex biological events such as domestication and/or husbandry practices.

The aim of the present study is to reveal diachronical differences between populations of Sus scrofa (pig ad wild boar), based on the analysis of hyper variable first part of mitochondrial control region. All the analyzed bones’ fragments and teeth are sampled from Poduri-Dealul Ghindaru Tell. The Tell of Poduri-Dealul Ghindaru (positioned in the Eastern part of Romania) has a complex stratigraphy and the archaeologists have carried out analyses on Chalcolithic (Precucuteni, Cucuteni) and Bronze Age levels.

The DNA template purification was performed using the DNA IQ Kit (Promega) for forensic analysis and quantified by agarose gel electrophoresis and spectrophotometery, PCR was carried out by using the Gotaq Green Master Mix (Promega) and two nested primers. Amplicons were tested by agarose gel electrophoresis, purified and direct sequenced with 8 capillaries CEQ 8000 Beckman Coulter analyzer. Sequences were aligned and compared with the haplotypes from databases and actual species and inferred for phylogenetic directions.
